                     I have one query regarding ligation of his tag sequence with unknown cDNA sequence that has to be cloned under prey vector for yeast one hybrid assay. How to make it possible??

The histidine tag I want to add with my cDNA  for screening the expressed protein nuclear complex through antibody against the tag amino acid sequence as epitope region.How it can be done to analyze through the technique-RIME protein immunoprecipitation??

Second:- Can you also please tell me some reliable kits name which are widely used for yeast one hybrid assay?
