I used Jasco FP-8300 spectrofluorometer. There is an option for using internal filters or not. I am sure these filters is used to avoid the effect of higher order diffraction of the grating. However, if the filter is used other problem came out, there will be broken spectra on two locations (there are three band pass filters used), please find kindly the attached image. The image is the light source spectrum image and it was taken using a diffuser to collect the light source characteristics of the spectrofluorometer. Please give me suggestion to use or not use the filter or on how to correct the results using the filters. I am also wondering how to check the effect of the second order diffraction of the grating.
