My plant extract is a solution & how to do the sodium hydroxide test to check for flavonoids?
Please state the amount of 10% NaOH, HCL and plant extract I should take for the test?
Do I need to warm and filter my plant extract solution in distilled water before adding NaOH?

Firstly, flavonoids are large group of compounds that are widely distributed in plants. In all cases, flavonids reacts with NaOH to give yellow colouration as the case maybe. Colour rate depends on the quantity of flavonoids it contains. Also, it is important to evaporate the extraction solvent to a minimum to avoid interference.
The amount of NaOH to use depends on whether you are caryingout only a qualitative or quantitative test. If qualitative test alone, 3:1 of extract and 10% NaOH will give good result. You may start by adding the NaOH in a drop wise manner.
Using a plant sample with established flavonoids presence such as green tea and the likes might help you reach a better conclusion of whether your extract actually contained flavonoids.

To the last part of your question, It is not necessary to warm your plant extract if you chose cold extraction method. However, you must filter your plant extract before the test proper.

5 gtts. of 5% NaOH was added to the extract followed by addition of 2mL 10% HCl.
A yellow solution that turns colorless upon addition of HCl indicates the presence of flavonoids.

I'd like to know what you're meaning with secondary solvents used in ethanolic plant extraction?. Do you mean solvents used to mix with ethanol?. If so, you should used
solvent miscible with ethanol, such as water; methanol, acetone, isopropanol, dioxane.

What I meant was after total evaporation of ethanol from the plant extract what are the solvents usually used or can be used to mix the plant extract for antimicrobial well diffusion inoculation?
Many papers have mentioned that they have used DMSO solution in ethanolic extraction like wise what are the other solvents that can be used?
Will isopropanol do?
