Try to determine the acid value of algal oil, to establish propper values of catalyst.  Also focus attention on alcohol quantity, reaction parameters like time, temperature or pressure.

I suggest to to do it through Gas Chromatography. It is sufficient to inject 1 microliter of your hexane solution containing FAMEs in the instrument after a caalibration done with an hexane solution containing standard FAMEs at a known concentration.

Devide Frumento I extracted lipid from dried algal powder and evaporated the solvent found dried lipid if I dissolved it in hexane and send for gcms then may I get lipid profile.

As Devide stated, FAME will be easily determined using GC. The detection metohod can be FID or a more sophisticated one, such as MS.

You can not easily identify the lipids by GC without derivatization into FAMEs, because the temperature will have to be quite higher for volatilization, and you will have too many isomers to have a good quantification. I see two options:

1) You can stick to transesterification into FAMEs followed by GC. No need to develop a transesterification method, there are several and many give similar results. I use acid catalysis with H2SO4. Be mindful of the water content in your reagents.

2) You can saponify the lipids and then convert them to fatty acids, and analyze these by LC-MS. If you use GC, again you will have to use high temperatures.
