I am working on anti-factor Xa activity assay. The absorbance values increase with decrease in the concentration, and the blank values are always higher than the rest of the concentration. How should I do blank correction? As I cannot subtract the absorbance value of blank from that of the concentrations is there any other way?
I don't know the assay but it sounds from the description that absorbance decreases with increasing concentration, i.e. decreases from the zero standard. In this case the appropriate blank (if any) relates to a sample with no activity.
Paul Milham Yes that's right, blank has no activity (it has no sample). Absorbance value of the blank is between 1.1-1.2. I start with the least concentration of the sample,and as the concentration of the sample increases the absorbance values keep reducing. I have to measure the absorbance of each concentration against blank. How should i apply blank correction? What am I supposed to do with the blank absorbance value?
Keyuri Mokashi , could you elaborate a bit more on how you perform that experiment? your blank absorbance already seems very high / too high to efficiently do absorbance measurements.
