I don't have advanced knowledge of SPSS and tutorials aren't making much sense to me, if anyone is able to explain SPSS two-way ONOVA step-by-step in the simplest form I would be grateful. My research is based on Raffinose and gut barrier function using TEER values. I have a control measure, high, medium and low dose over a time frame of 0,4,24 and 48 hours. How do I input the data and analyse into a two-way ONOVA?

It sounds as if the dose factor is a between-subjects factor (4 levels: none, low, medium, high) and the time factor is a within-subjects or repeated measures factor with 4 levels (0, 4, 24, and 48 hours). That's a pretty common anova design, variously referred to as: split-plot / mixed / one-between, one-within model.
In spss, the data entry would require 5 columns per case: one to indicate dosage level, and four for each of the observation/measurement time results.
The analysis can be elicited by clicking (from the main menu bar): Analyze / General Linear Model / Repeated measures...
In the dialog box that follows, name your repeated measures factor (e.g., Time), and enter the number of levels for that factor (4). Click "add" then click "define" at the bottom. The main dialog box then pops up. You'll enter your four time measures as the Within-subjects Variables (in order), and enter your dose factor as the Between-subjects Factor.
If you want descriptive stats, and other goodies, explore the "Options" button. As well, explore the "EM Means" button if you'd like to see follow-up tests on the time dimension levels.
