How to distinguish between endolichen fungi and mycobiont isolated from unidentified lichen?

Lets assume, A number of fungi were isolated from the thalli of an unidentified lichen. Later it was confirmed individual fungus by means of ITS BLAST, but, how to know which fungi is mycobiont among those fungi?
