I am not sure whether this is possible with purified ATP synthase, but treating isolated mitochondria with relatively high concentrations of calcium can detach the F1 unit from the remaining ATP synthase. Please see attached paper.
Thank you for your answer, @Gisela. My aim is disassemble the F1 complex into individual subunits, but I didn't find any reference has done that. I don't purify the complex but treat the Mt lysate.
Could you treat isolated F1 subunits with RIPA buffer or any other buffer that contains a low concentration of SDS and perhaps dithiothreitol? If using isolated mitochondria you may need to immunoprecipitated the F1 subunits from the lysate.
