I wanted to analyze total phenolic content in plant extracts using Folin-Ciocalteu method. In order to get calibration curve I used gallic acid. But after adding all reagents (for standards and samples) white colour sediment was observed. I tried keeping the tubes in the room temperature for 30 min and also keeping in boiling water bath for 1 min. But both methods produced white sediment. Please can anyone suggest me haw can I solve this sediment problem?

How do you prepare the plant extract? We have worked with different solvents (water, ethanol, methanol...) and different vegetables and maybe the problem is the extract concentration, sometimes we had to evaporate the solvent in order to concentrate a lot the sample.
Thank you all. We used ethanol and methanol (80%) to extract vegetables. Yes what you said is correct. We repeated extraction again using the same solvent and evaporated fully. Then it worked and we got proper results. Thank you all again.
