I would like to know how to determine the amount of functional groups, here carboxyl groups, in PAA (100 kDa). I would guess by acid-base titration.

In detail I would prepare a NaOH standard solution (~ 0.05 M) with potassium biphthalate as a standard. Furthermore I would prepare a HCl standard solution (~ 0.05 M) also with potassium biphthalate. The polymer would be dissolved in the NaOH standard solution. After that the excess NaOH in the solution would be titrated with HCl standard solution with phenolphthalein as an indicator.

The reason is, that I would like to repeat the same experiment with my hydrophobically modified PAA (HM-PAA) in order to know the amount of carboxyl groups before and after the reaction and the fraction of modification. Apart from that I would like to control the deprotonation of my HM-PAA in order to make it water soluble. An example: How much NaOH do I have to add in order to deprotonate 10% of HM-PAAs' carboxyl groups.

Is it possible to proceed according to the mentioned procedure and which polymer concentration shall I use when dissolving.
