There are different methods to identify the promoter sequence of a given gene.

1) You could use a prediction tool. Please have a look at this collection:

2) Cloning putative promoter sequences in front of a reporter gene is one way of validating the prediction. Small mutations in the promoter sequence could be used to identify important elements.

The first method is easy, but not as reliable as the second. Do you need detailed information about the promoter sequence or would it be sufficient to use several bases upstream of your first gene set? Screening both gene sets with the before mentioned tools could reveal internal promoters. However, you should be aware of the high number of false positive hits these tools could report.

will look into the provided online prediction tool. However, I am primarily interested in the expression of both of the genes rather than promoter activity. I just want to express them in heterologous expression system such as pET system. So, just want to know whether both of the genes should be expressed together or they can be expressed individually followed by in vitro reconstitution ?
