I'm designing CAR-T cells targeting solid tumors. But I was stumped entirely by the designing of the gene sequences for CAR-T. I want to design anti-CEA and anti-MLSN CAR-T cells. I can find the gene sequences of them but I don't know how to choose. I found some patents of CAR-T cells, but some intracellular regions such as CD28 and 4-1BB. Even if the same molecule they used, the gene sequences are different. Some of them just have several different amino acids, some have a lot. I want to know the steps to choose the sequences of anti-CEA and anti-MLSN scFv and the intracellular regions for CAR-T cells. And how could I make sure the gene sequence? I hope for the reply.
