How to Design a probe?

Well, I am really confused with this probe thing. For example, if I am looking for an unknown gene in a long stretch of DNA using Southern blot technique,how would I design a probe without knowing the sequence of the gene?

Even in DNA fingerprinting, how can I design a VNTR probe without knowing the DNA fragments base sequence. Please help...
