I need to design a sandwich lateral flow immuno-assay to detect two concentration ranges of the same antigen i.e. one test line can detect lower concentration of antigen (1500pg). I am struggling with choosing the right antibodies for this test.
My current idea is to use AuNP coated one antibody at Conjugate pad, Test line 1- low concentration of 2nd Ab to capture low amount of antigen, Test line 2- High concentration of Ab to capture remaining amount of Ag, and Control line- IgG....
Is this a correct set up? If not why?
I think it is fine with your set up, your just need to play with the sensitivity you desire which will be depending on how good of your antibodies are in terms of affinity & avidity and either they're mAbs or pAbs. As you said, for example, the 1st test line sprayed with low conc of capture Ab will detect higher LOD, (>1500pg/mL), whereas the 2nd test line with high conc. of capture Ab will detect lower LOD (
