I'm trying to prepare 5% Tween 20 in cell lysis buffer, if I pipette the tween 20 directly from the stock is stuck in the tip (as it is viscous) & if I heat the tween 20 stock in a water bath I increase the probability of oxidate the tween 20 (as it is sensitive to the light & heat).
SO, what should I do?

For preparing 5% Tween 20 in cell lysis buffer, instead of using tip and pipetman, try to change the volume to weight (density of tween 20 is 1.1 g/mL) and follow these stages:
1. Weigh the required amount of tween 20 in a glass beaker.
2. Heat the beaker on the hot plate stirrer at 40-45 ˚C.
3.Add the buffer solution to the beaker slowly while its contents are being stirred.
I've worked with tween 20 previously in this way:
Use a 1-ml pipette tip because it has a wider orifice compared to the 200-ul tip. Aspirate the calculated volume that you need to transfer. When you dispense, make the pipette tip orifice below the liquid surface, after completely dispensing, re-aspirate from the liquid and dispense many times (as that you are rinsing the inside of the pipette tip). In this way, the highly viscous tween 20 in the tip is all transferred to the solution.

I expect that you encounter these problems due to the yield stress of your solution. There are a few simple directions that you can apply.
Use a pipette with a larger inner radius or a shorter length, so you can more easily overcome the yield stress.

Is your question: (Does mixing tween 20 with other compounds in a bigger solution decrease tween 20's ability to react as we would like it to do so?) If this is your question, the answer is: it depends. I was diluting tween 20 in water which shouldn't affect its reactivity potential. If there are other compounds (methanol, acetonitrile, etc.) in addition to water, then it may reduce the reactivity of tween 20.

I would like to consult you about some technical issues regarding Tween 20 usage. We are using Sigma-Tween 20 EMPROVE ESSENTIAL for dissolution studies of our product. The product is a form of nanosuspension and the molecule in the suspension has a low water solubility so it is hydrophobic. We think that because of Tween 20 molecules having tendecy to adsorb at the hydrophobic molecule-water interface, the results we obtained are not consistent. Is there any solution for this problem? Is there any other kind of Tween 20 you could recommend besides EMPROVE ESSENTIAL grade? Is there a way you could suggest during the preparation of the dissolution medium contains Tween 20 so that Tween 20 is not being adsorbed on hydrophobic molecule? How HLB and CMC values effect the adsoption of Tween 20 molecule on hydrophobic molecule?
