How to conduct protein extraction of secreted ECM proteins from soft agar assays?

Hello, I am planning soft agar assay with MCF-7 cells. The cells will be transduced with an adenoviral vector before the assays and then cultured on soft agar. We are studying one of the secreted ECM proteoglycans. Is it possible to extract secreted extracellular proteins from the agar?
