In the Abstract, it states"Furthermore, we developed a biolistic method for protein delivery based on the known proteolistics technique." Take a look what they did in the Method section. Look like pretty easy. You are using Cas9-GFP protein (without gRNA linked), and they used Cas9-GFP/gRNA.
In the paper:
" We also tested whether biolistic bombardment could be used to deliver the Cas9-GFP/gRNA RNP into non-transgenic BY2 cells. In contrast to the negative controls containing the W5 buffer and the gRNA (without the Cas9-GFP fusion protein), we found that weak positive GFP signals could be detected on the membranes of some bombarded BY2 cells at 6 hpt (Fig. 4). At 48 hpt, strong GFP signals were detected in the nuclei of some bombarded cells with decreased GFP signals on their membranes (Fig. 4). These results indicated that at least a portion of the preformed Cas9-GFP/gRNA RNP complex retained its integrity, when bombarded into the BY2 cells and these intact RNP complex molecules had been successfully intracellularly delivered into the BY2 nuclei. Therefore, biolistic bombardment would be an effective method to deliver the Cas9-GFP/gRNA RNP into BY2 cells.
