1. Can anyone suggest me the meaning of "fungus has lost its virulence"?
2. If I am able to observe micro and macroconidia under microscope, does that mean the fungus is still virulent?
3. Also, how to check the virulence without bioassay?

I will try to answer within my knowledge
1. Depending on the species, the degree of virulence is greater or less. If it is an opportunistic infection fungus, the degree is limited - the fungus grows in a host that has low resistance.
2. I believe so. I don't know much about fungi morphological changes associated with virulence. I think it is something intrinsic.
3. Database. This part includes bioinformatics. There are certainly articles that show the sequencing of genes that cause the virulence of certain fungi. With these sequences it is possible to make an alignment and identify where there are changes that may or may not change the degree of virulence.
