I am new in working with Diva and FlowJo and I need help with a technical issue. During my analysis with Diva my voltage settings of FSC and SSC were correct because I could see my population of lymphocytes in the graph. But now I am analysing my data with FlowJo and there is a weird line instead of my population of lymphocytes. Does anyone have an idea how to fix this?

Editing my answer as I realized I had stained cells in mind. Increase the scale on FSC-A until you see clear populations. If I remember correctly, you click on the T button right next to your FSC-A and then there should be aan option where you can change the scale. If you are looking at stained cells, use log scale instead of linear. You could use biexponential scale instead if you are losing populations on the left.
Dear Cheyenne, you cannot change the voltages on FlowJo. That’s why you have to check during the acquisition that everything is ok. In your case it was, so it should be fine. When you see this kind of line this means that it is a scale problem In FlowJo. All the events that were « stuck » on the right side of the FSC axis during the acquisition (saturating in FSC) appear in this line on FlowJo. To change the vizualization just click on the T button on the FSC axis, let it in the linear mode (never put FSC and SSC in log) and try to play with the + and - buttons under the graph that you see. It will allow you to put your scale in the right way. Hope this helps
