I usually calculate using the formula c1v1=c2v2 but have observed other people calculate differently. It confuses me.

For example, if the number of cells to be seeded is 1x10^4 and the stock is 86.5x10^4/ml, they just divide 1x10^4 by 86.5x10^4. Is this correct? I mean, the stock value is just for 1ml and not the total amount in the tube.

Another problem is when calculating the amount of DNA. If the given stock of DNA is 15ng/ul and the required amount of DNA is 40ng, they divide 40 by 15. The total volume of the reaction mixture is 25ul but why is this volume never taken into consideration? Why can't I use c1v1=c2v2?

Can somebody please help me clear this confusion?
