How to calculate substrate amount, kinase amount and reaction volume, if the activity of kinase is 16nm/min/mg?

I am doing in vitro reaction with recombinant substrate protein and kinase. We got the activity of the kinase, 16nmol/min/mg, how much substrate protein and kinase should I use? And how much should the total volume of reaction be?
