can anyone tell me the calculation for MTT assay. Iam going to use triplicates of my sample and control in a microplate reader. I have untreated and treated samples. Do I have to have a negative control (MTT without cells). help needed.

MTT assay is a Comparative Test that you compare a untreated sample with one or more treated sample(s)
you should read concentration of furmazan in each sample and then depending on your samples ( exp in cancer cells you want to have less cell than untreated cell ) you should divide concentration of treated cell on untreated cell and calculate the percentage of mortality in treated samples.
