I have injected 5 samples of lowest possible concentrations. I have taken the std deviation of area and slope of the equation obtained for the regression line of calibration to calculate LOD & LOQ using formula.

But, in the Reference Book viz., "Analytical Method Validation and Instrument Performance Verification" by C. Chan, et al. it is suggested to take the residual standard deviation of regression line of calibration curve to calculate LOD & LOQ. So, I am confused how to calculate the residual std deviation? Also I want guidance as to whether we are supposed to take std deviation of area or y-intercept by performing same experiment in sets?
