Currently I am working with my plant extracts with different concentration on DPPH assay and anti-tyrosinase assay. All of my plant extracts with different concentration are dissolved in DMSO. I would like to ask do I need to subtract the inhibition percentage of DMSO only from the inhibition percentage of DMSO + plant extract in order to eliminate the inhibition by DMSO solely when I am calculating the IC50 of assay
*(Inhibition % of DMSO + plant extract) - (Inhibition % of DMSO) = (Inhibition % of plant extract). Then use the (Inhibition % of plant extract) calculated to generate IC50 graph in Microsoft excel.

You will find it best to determine the concentration of DMSO that doesn't cause interference with your assay. While DMSO may not bind to your substrate in a competitive fashion, it will solvate both the substrate and the compound being studied and will prevent binding.
@Zhou, usually DMSO is safe for IC50 determination when you use that to dissolve/ suspend the sample. Since you will use spectroscopic technique (preferably UV), the transparency of the mixture is not affected at the desired lamdba. So just calculate as it is, dont use your new formula.
Hi Dr Aziz Rahman, but the presence of DMSO solely will have the inhibition effect on my assay. So can I just neglected the result of DMSO solely?
