How to calculate ED50?

Ive been working with a peptide and an algae extract and for in-vivo part of the project i have to treat them as an ointment on excision wounds on rat models. so i need to calculate the ED50 of them to apply on rats .Can anybody share the experience about this issue with me or suggest a way about how to convert the concentrations to ED50?
