How can one calculate the copy number with following details:

plasmid size 4000 bp

concentration 1600 ng/ul

Now I did try using thermoscientific online tool which does tell u total copy numbers per ul with above details.

but I find the tool is misleading the calculation.

for example: using above values the tool gives me the number of copies per ul right. (eg: if it says 3X10^11 copies/ul)

Now if I wish to make a copy number of 50 copies/ul in 10 ul it shows that I need to add ~1.2 ul + 9.9 ul , now If i raise my copy number to 500 copies/ul in 10 ul it says the same 1.2+9.9? how.?

Other strategy which I guess most people follow : from the main stock copy number calculate it for 1X10^8 and then make serial dilutions to bring it to 5 copy number?

Which is the better option?

Application is to use the plasmid copy numbers for standard curve preparation to determine viral dose in clinical samples.
