When I use the protocol to digest the slice of tissue some holes can be viewed. How can I improve my trypsin digestion protocol for not to occur that?
The deposition of trypsin is done with the aid of Bruker's ImagePrep and the incubation was done for 16 or 3 hours at 37 Celsius degrees in a humid chamber.

Thanks for the suggestion , but I believe this is not the case, because the holes only appeared after the deposition of Trypsin. In the other slides of the same series, that I did not use Trypsin, I was able to do the analyzes without these holes appearing.

Hi Felipe,
It sounds like you might be having issues with uneven matrix deposition. Bruker has discontinued the ImagePrep and has switched to using the TM-Sprayer for MALDI matrix deposition. I would suggest looking at this product.
