I'm trying to quantify the percentage of actively dividing yeast cells under many conditions, including cells treated with alpha factor. I was planning on taking phase contrast micrographs of my cells and feeding those pictures into one of the many image analysis software packages available (i.e. Cell-ID or CalMorph). Would it be difficult for these programs to differentiate between budding and shmooing cells? I am open to other methods, as well. For those who do this sort of work, what is your preferred method? If you use an image analysis software, which is your favorite? Any help would be appreciated!
