In my work, I need to attach gold nanoparticles (modified with biotin labeled dna) to streptavidin coated polystyrene beads.
However, it didn't work well in experiment. In my knowledge, streptavidin and biotin have very strong affinity, I don't know why the binding cannot happen.
Is it possible because both gold nanoparticles and streptavidin coated polystyrene beads are negtively charged, so they repel each other? Is there any solution to solve this?
Any comment will be sincerely appreciated.
Did you confirm whether both particles are negatively charged through, e.g., Zeta potential measurements? Since the net charge depends on the pKa in relation to the pH, are there options to change the pH?
Alternatively (at least if this is compatible with your research), you could opt for a binding between gold and thiols instead.
