I need to attach a protein (carrying a N-terminal cysteine residue) to spherical gold nanoparticles in a defined ratio. Due to recombinant expression the target protein carries a HisTag as well. My hypothesis is that both (cysteine and histidine) could bind, but the thiol-based cysteine reaction binds stronger and with higher affinity. How can I support gold-thiol binding even more, any ideas?

Another problem is the subsequent passivation with thiol-PEG which competes with the protein binding. Does someone have an idea how to avoid replacement of the protein?
