I am looking to analyze ARB and ARG from real surface water of marshland.
What you should be more concern with is how to isolate the bacteria from the water sources. Analyzing ARB or ARG is done similar to that of clinical sources
You can consider the utilization of eDNA if you want to fully assess the ARGs present in the surface water of Marshland. Focusing on the Marshland alone might limit your analysis. You can also consider analyzing the nearby water sources such as rivers and waste water facilities that might have a greater impact in the increasing problems on ARGs. Good luck with your studies.
First step:
Collect water samples
Isolate and identify bacteria present in the water samples
Subject the bacteria gotten to different antibiotics
Pick those bacteria that are multi-resistance
Pass them to plasmid analysis
Select primer that has the resistance genes, do the PCR on those one with plasmid and Multi resistance,good luck
