I have a drug that is non-polar in nature (dissolved in DMSO or acetone), but I would like it to be incorporated in my internal solution and my bath (which is polar in nature). I was wondering whether anyone can give me a hand please.

You can add a few drops of DMSO in the eppendorf of your internal solution, and same can be done in the external (just use a bit more), but careful because DMSO is directly affecting cell viability through lipidic membrane dissolving, so use just a bit. I would say 2 or 3 ul in the internal solution per eppendorf.

you can add your drug dissolved in DMSO directly to your solution. Up to 0.5% v/v of DMSO can be added to the internal or bath solution. But, as Marcel said, DMSO reduces cell viability and makes patch clamping more difficult.

You could consider employing solubilising additives to your solution, such as aminoacids, PEG, etc.
