I am looking for a standardized method which can be used for plasmid curing in E.coli. Any information on tried and tested methods would be of great help.
I work on Ethedium bromide, Vitamin C, Acreden orange and many detergents its use to curing plasmid in G-ve specially E.coli, and do gradient concentration about this materials and see in which cons. curing plasmids
no standard methods exist for plasmid curing because of different mechanisms of replication and maintenance in cells. A simple and efficient way, tested by myself, is treatment with low concentrations of SDS. It worked well to cure stable plasmids like IncP group members. Good results were also obtained with ColE1-derived plasmids using mitomycin C; attention, it is carcinogen!
