I’m preparing LY294002 (HY-10108) for in vivo use in mice (10% DMSO + 40% PEG300 + 5% Tween-80 + 45% saline). At 50 mg/ml in DMSO, the solution isn’t fully clear, and at 5 mg/ml working concentration it often turns turbid. Should these solutions be completely clear, and is it safe to inject with 10% DMSO? Also, is it better to prepare fresh working solutions each time or can they be stored?
During your entire experimental process, you need to pay attention to the following:
1. The DMSO mother solution of 50 mg/ml is not completely clear, which should not be the case. We measured that 50 mg/ml can be completely clear. In the subsequent step, it is recommended that you assist with ultrasonic heating to dissolve and ensure complete dissolution.
2. You can't use up the system for dissolving 50 mg of drug at one time? For the working solution, we recommend that it be prepared and used immediately, and it is not recommended to store it because the system is too complicated. For the DMSO stock solution (50 mg/ml), it can be stored in subpackages. When using, take out a tube and add PEG300 or other co-solvents to dilute the drug.
3. If the working solution of 5 mg/ml is turbid, you can assist with ultrasonic heating to dissolve and see if a uniform suspension can be achieved. The uniform suspension can be used for intraperitoneal administration.
4. In this solution, DMSO accounts for 10%, which is tolerable for normal mice. If the mice are weak or have been modeled, it is recommended to consider the ready-to-use solution: the last solution on the official website is 0.5% CMC-Na/saline water.
5. Ultrasonic cleaning instrument，20-40 kHz, temperature 37-50 ℃.
Ultrasonic time: 3-5 min each time, repeat several times, and observe the state of the solution.
