During animal cell subculture,After estimating the viable cells using automated cell counter machine, how researchers know that this much amount of cells to be added to this much amount of growth media so that it proliferates and attain 80% confluency within 48hrs?
I want to know the calculation part.

The cell number completely depends on the cell type you are working with. The number of cells we seed for highly dividing cells are different to that of slowly diving cells. This number is important if you are doing stem cells culture. But basically we usually perform the seeding density analysis of the cells and look for the morphological characters and other characters to determine the best seeding density. For this analysis we seed the different number of cells and analyzed the cells in different time point.

I agree with the other member's answers. It totally depends on the cell type in which you are working, cells doubling time, seeding density, and most important tissue culture plate size.
