I have the following conditions:
-perfect amplification
-a 3000 bp amplicon
-a %GC content of exactly 50%
-15 cycles
-0.5 mM dNTPs (A, G, C, T)
-0.5 uM primers
-10 ul reaction volumes
I need to know how much of the reagents are consumed after 30 cycles?
I do not think that you can answer this question because you have not specified how many copies of your template are present at the start of the reaction
Also, you can't estimate how much of the dNTPs are used because of the presence of the longer than your desired product amplification that occurs in the first few cycles. You could, I suppose ignore that part.
Can I ask why you need to know this?
Usually PCR Master Mix (MM) will have an excess of dNTP's as it will be a limitation factor for amplification, same with the primers. In most cases the conc's of primer, probe and MM are known and if not using the checker-board approach to figure out the best ratios is the way to go
