Hi I am reviewing protocols for plant media in the lab I just started at and I am wondering how much MES Hydrate should be added to the media. Each protocol is different. some split the MES in half for 500 mL others (for the same media) call for 0.5 grams still. is it 0.5 grams per liter or is it always 0.5 grams. I am just trying to set constant standards. Thank you.
MES buffer is a Good's buffer that is remarkably stable both chemically and enzymatically. It has minimal UV absorbance and is often used as a running buffer for bis-tris gels. Prepare 800 mL of dH2O in a suitable container. Add 97.62 g of MES free acid to the solution.
0.1 M MeS Buffer: Dissolve 19.2g of MeS free acid (MW 195.2) in ~900ml of pure water. Titrate to desired ph (5.2-6.0) with 1n naOh. Make up volume to 1,000ml with pure water. 2% carbodiimide: 2% 1-(3-Dimethylaminopropyl)-3-ethyl carbodiimide hydochloride dissolved in MeS Buffer.
MES is often used in buffered culture media for bacteria, yeast, and mammalian cells. It is toxic towards plants at high concentrations, but can be used in culture media at lower concentrations for plants. Due to low ionic mobility, MES is regarded as an excellent buffer for use in capillary electrochromatography.
