1-3 mil, depending on the accuracy of your method

Sadly its a very small number! To get usable amounts I would suggest you pool several mice or flow whole blood to avoid massive loss due to the buffy-coat step. It works relatively well! Sorry I don't have better news.

This strongly depends on the donor / source.

I am characterize them by flow cytometry using multiple panel (CD45, CD11b, Gr1, CD3, CD4, or add CD34 etc.), depending of interest.
