I'm curious if anyone knows how long Brefeldin A is stable for at 4 degrees centigrade?
Our current batch is 5 years out of date and I would like to buy a new batch, but my boss thinks the brefaldin is stable and it should still be usable.
Has anyone had any experience in Brefeldin A going off?
Anthony, 5 years out of date is a bit long. We used Brefeldin A that was 2 years out of date, definitively didn't do the job as well. So, it definitively goes of. Hope that helps.
Hi, in case you need to confirm, if you have antibody against any golgi membrane protein (eg, giantin) then you can quickly test the functionality of BFA. About 30min incubation to cells (in the range of 1-10µg/ml) should homogenously disperse the golgi. If not, then its definitely not working.
If the stuff is still in crystal form, I think it should be fine. Perform a quick intracellular cytokine assay with T cells or NK, you will find that out. Alternatively, infect a cell with a virus in the presence or absence of your BFA (10ug/ml final concentration) overnight, then stain for cell surface viral antigen. If no staining in the plus BFA group, then it is working fine
