I'm looking for some research (article) about Agrobacterium tumefaciens live shelf at the plant cells after infection? After infection, the ti-DNA is transferred to the plant cell and genes are integrated into plant genome, so what is happen then to the bacteria? Are they alive? Can I found they if I will growth plant tissue on semi-selective media. If anybody has some experience with it?
i am not clearly for this question, but interesting in . maybe its alive after the t-DNA has been transformed into g-DNA at starting , but for longer regeneration, is the vectors alive along ? i dont know
Do you think after the insertion of T-DNA into host cell, the bacteria are still alive or virulent?
At least, I can say that:
1) Agrobacterium rhizogenes is present in vascular system of infected plants, and even can be found in fruits and seeds
2) Inner tissue of crown gall (A. tumefaciens) of large size is usually free of the pathogen, but can be colonized by other bacterial species (we found Erwinia, Pantoea, Staphylococcus, Pseudomonas, etc.).
3) Agrobacterium spp. population multiplies around the infected plant using opines synthesized by the tumor tissues.
4) A number of infective bacteria around the target plant cell is large so we can assume that they stay alive and virulent for long time.
I do not think that the bacterial cell that infects plant cell gives away its very last copy of Ti/Ri-plasmid.
thanks for your answer , Mr. Alex , Do you know the Agrobacterium spp could inner the infected plant cell or the only T-DNA fragment could insert into g-DNA ?As you said that the Agrobacterium is present in vascular system almost -- understand for me from this sentence is the bacterium do not into the infected plant cell but located in the intercellular . is it right ?

Regarding the A. tumefasiens, as I know, the bacteria are present within the plant not systemically. 20 cm upstream from the soil surface you will not detect any bacteria in the plant tissue. The wound will be closed by callus formation after 7-10 days and there is no bacteria entrance, otherwise there is a new wound and a new infection. The question is after the T-DNA insertion, are the bacteria viable or not? From the diagnostic point of view, if I will grow some plant tissue on growth media, will the agro grow? Or should I test for T-DNA genes by PCR?

Usually, we kill Agrobacterium in transformed cells by Cefotaximum 500 mg/ml (Roussel Uclaf, France) or other antibiotic treatment for 4 weeks.
and test resulted plant tissue by PCR.

From reading your discussion above, it sounds like Agrobacterium tumefaciens is not systemically found throughout an infected plant’s tissue, but localized to tissue surrounding the wound/infection site. Is this true? Thank you! Hila Shomin Alex Ignatov
Jennifer Koym At least, it is rare found in crown gall tissue at late stage of infection. Some data about A. bv1 "crazy roots" infection:
"The percentages of Rhizobiales clones (including Agrobacterium spp.) were greater in the rhizosphere of plants grown on rock wool mats (infected (by Agrobacterium) - 25.9% and control -26.1%) than on coco fiber mats (6.6 and 8.1%). ... Agrobacterium spp. was a predominant part of Rhizobiales clones in infected rock wool mats (72.7%), while infected coco fiber mats had only 32% of Agrobacterium spp. in total number of Rhizobiales clones.
