In my study of biomass pre-treatment, I have carried out acid hydrolysis using Sulfuric acid. After separating the hydrolysate, I am checking the amount of fermentable sugar released. As from the literature, acid pre-treatment works on only the hemicellulose fraction. But can there be glucose monomer too ??
To have clear account of the amount of sugar released.
1. Orcinol method for only pentose sugar released (as suggested to other Research gate followers)
2. DNS method for Total reducing sugar. (absorbance at 540 nm against standard Glucose)
How can I have an idea of the efficiency of the treatment, in terms of pentose and hexose (if any ??) sugar yield.
