I think the best way to justify, is by carrying out hydrolysis for both dry and wet biomass for a few hours and measure the sugar content in the end.
Hello Mam, In this perticular case porosity of material played major role in limiting enzymatic hydrolysis. So in my point of view any method or instrument which can detect porosity of both wet and dry material will be helpful.

Thanks Amit for your valuable inputs. Its very difficult of measure the true values of crystallinity, porosity or pore size distribution while dealing with the wet substrate. All these parameters eventually require drying and once dried all the parameters are represent the dry biomass and not the wet one.
