The way how platelet lysates, in general, and PRF, in particular, can reduce pain and inflammation is not fully understood. A limited number of publications have highlighted the role of TGF-b and HGF, released by platelets, in inhibiting some inflammatory pathway leading to reduced production of inflammatory cytokines like TNF-alpha. More research is needed to understand this anti-inflammatory/analgesic role especially as it is likely to vary depending upon characteristics of the platelet materials and the tissues. I can only hypothyse the following: Previous studies have associated IL-1B, TNF-α, IL-6 and IL-8 with pain. The release of IL-4, an anti-inflammatory cytokines found in PRF, modulates inflammation and pain by inhibiting MMPs and neutralizing transduction pathways from IL-1B, TNF-α and prostaglandins.
