Does a single phosphorylation site have an effect on protein mobility on SDS-PAGE gel?is that due to change in size, or charge? Thank you all for your thoughts :)
Charge should mostly be irrelevant in SDS-PAGE because the point of the SDS is to neutralize the charges, denature the protein and have the proteins run based primarily on mass. The size difference of one extra phosphate would be undetectable (the mass of P is 94, the mass of an average protein is 40,000). So in most cases you would not see a change.
phosphorylation add negative charges to proteins which effect their functional properties , and effect the proteins mobility on alkaline PAGE , but in the SDS-PAGE the effect of phosphorylation is low because treatment with SDS will convert all protein charges to negative ,
