I believe they should only have an effect on the diffusion distance and in controlling flow velocity or keeping Reynolds no in the Stokes flow regime. Is there any other significance to this parameter in assay design.

There are many different immunoassay formats possible. First, you have to define the assay (with parameters) you want to perform. Then you can decide, which properties are necessary for your microchannel. There is no "universal reaction chamber".

I want to use microspheres attached to primary antibodies as a reagent, and then measure the optical signal in the chamber arising due to this binding.
