I'm interested in the production of insulin at a lab-scale. I read some papers about the topic. One of them mentioned autocleavage of the proinsulin without any enzymatic cleavage. But they didn't give any references or details about that. I sent emails to them but they didn't write anything. I just wonder how does autocleavage of proinsulin occurs? Could you help me with that, please? Are there any references or protocols about that?

Hi Esra,
Autocleavage can happen if the protein is an enzyme. In pancreatic beta cells, proinsulin is generally cleaved by PC1/3 enzymes to yield insulin (by removal of C-peptide). Proinsulin might also be cleaved non specifically by furin in some cases. I am unaware of proinsulin autocleavage. Would you mind sharing those articles that talk about autocleavage of proinsulin?

I could not find the "autocleavage and proinsulin" in that reference, unfortunately.
Also, I would like to ask a question, I use the trypsin, enterokinase, carboxypeptidase etc. but I didn't hear any PC1/3 or furin. Are they better? could you share some information about that, please?

Thanks for sharing! But this paper provides a poor citation of the autocatalytic cleavage part; Gooch 2011 that only gives you the definition of "autocatalytic" and nothing else! I would not follow this, and still continue doing enzyme based cleavage. I would be interested to know if you hear back from the authors providing some explanation or protocol for this.
