Inactivation and Treatment of sera to remove nonspecific haemagglutinins
Sera is treated with (RDE) receptor destroying enzyme from Vibrio Cholera, performed in a Biosafety cabinet, add 50 µl of serum to 200 µl of RDE, incubate overnight (12-18 hours) in a water bath at 37 ° C, add 150 µl of sodium citrate solution 2.5% and inactivate by heat for 30 minutes at 56 ° C. Mix 200 µl of treated serum with 25 µl PBS, and then add 50 µl of male turkey erythrocytes 50% to give a final serum dilution of 1:10. Stirred and incubated 30 minutes at room temperature.
