I am working on pollen storage of some fruit crops. I will extract pollens from anthers and then will store them in some organic solvents. Can anybody suggest some methodology for extraction of pollens from anthers and then isolation of pollens from organic solvent.
I am planning to use Grade 5 Qualitative Filter Paper Standard Grade fro isolation of pollens from chemical. Will this work?

You can email Dr. Sheila McCormick (from Plant Gene Expression Center, and professor of UC-Berkeley). She is also a member of ResearchGate: ( https://www.researchgate.net/profile/Sheila_McCormick ). Her lab is dealing with all aspects of plant pollens. I have seen her students using a home-made hand-held vacuum device to collect the pollens from the Arabidopsis plants in the greenhouse. I would like to know why are you using solvents to store pollen?
Last year i used to collect and store pollinia of orchid in UK. The onnly concer is how long they can stay viable. Some drying over silica (white with orange inidcator) may help. I think with they could be maintained dry and cold they will store well.

Why are you using solvents to store pollen. If you are going to loose the germination properties anyway I will suggest you to use something much cheaper like normal fixation with alcool or acetic acid. If you do intend to preserve viability, then I think you should use low temperatures. Get in touch if you want to discuss further, I work with pollen in Brazil so we can share experiences... Cheers
