I am trying to establish a cell line based xenograft model of CML in SCID mice. I tried to inject 5x10^5 K562 cells (in saline for injection) but there was considerable difficulty in injecting the cells via the tail vein (used an insulin syringe). There was mainly back flow of the cells. I tried in another animal, but this had gone in subcutaneously. 

Can anyone suggest any specific method they have used to deliver cells via the tail vein?
