I'm currently working on screening the efficacy of some novel drug with the cell line HEK293t. I have set of data for each drug at different concentration ranging from 0, 6.25mM, 12.5mM, 25mM, 50mM, and 100mM. I calculated the number of dead cells after propidium Iodiode assay and the values are normalized. I have also plotted the graph in Excel with concentration on X-axis and Mortality on Y-axis. I'm quite confused and stuck in identifying the IC50 values. Can you please help me to sort of this maths?

IC50 being the concentration providing half of the maximal effect, you can roughly estimate it on the graph (if data are "nice").
Otherwise, you can compute IC50 after fitting your data with a specific model : Emax-inhibition, or logistic (depending on the parametrization).
You need a specific software (user-made macro exist in excel).
I can help on one data set if needed.
If the effect is linear and maximal concentration is exceeding the IC50 (e.g number of viable cells) you are going to be able to calculate the value as is suggested previously. However if you dont have ''nice'' results you cant do it...
You can also try the BMDS software. That should do the work. See
